Principle Procedure Reagent Result and Preparation Urea Spot Test

The urea Spot Test is important Quality test used to detect the presence if urea in a simple. This test in common test in Biochemistry and Industrial use for Student During the Practical exam abd basic Analysis. The test is based reaction of Urea Dimethyl Amino Benzaldehyde (DAMB) Reagent producess a Yellow Colour Urea Persent.

To Detect the presence of Urea in the Collected Sample Using Reagent DMAB (Dimethyl Amino Benzaldehyde).

Principle Of Urea Spot test

Urea react with Dimethyl Amino Benzaldehyde in Acidic medium to form Yellow Color. Appear Yellow color Confirm the presence of Urea in the sample.

DMAB (Dimethyl Amino Benzaldehyde) Reagent Preparation

  • 1.6gram DMAB (Dimethyl Amino Benzaldehyde) in 100ml of acid alcohol
  • Add 10ml Concentrated HCL

Need Of Chemical and Requirements

  • About 1 Gram Sample
  • Charcoal 2-3gram
  • Distilled Water
  • Whatsman Filter Paper
  • Teast Tubes
  • DMAB (Dimethyl Amino Benzaldehyde).
  • Cyclo mixture

Procedure Of Urea Spot Test

  1. Take About 1gram of Sample And 2-3 gram of Charcoal in test Tube.
  2. Add Some Distilled Water For Dissolved.
  3. Mixed the Solution with cyclo mixer.
  4. Filter the solution through whatsman filter paper another test tube.
  5. Add 4-5 Drops of DMAB (Dimethyl Amino Benzaldehyde) Reagent Filter Solution.
  6. Yellow Color Indicates Urea Positive.

Note- Use Fresh DMAB (Dimethyl Amino Benzaldehyde), Handle Conc. HCL Carefully, Clean Test Tubes and Filter Paper, Not excess Reagent.
